    Ten zuiden van het Piazza dei Priori ligt Piazza San Giovanni, het religieuze centrum van Volterra. Het plein is bekroond met de 12e-eeuwse Duomo, met zijn Pisaans-Romaanse gevel. De kathedraal, die officieel bekend staat als de Cattedrale di Santa Maria Assunta, is op dit moment helaas gesloten wegens restauratie, waarschijnlijk tot september 2019. Binnenin zijn de hoogtepunten het indrukwekkende cassetteplafond en een verzameling aan kunstwerken daterend uit de middeleeuwen tot aan de renaissance. Het beeldhouwwerk van de Afzetting is een extreem zeldzaam Romeins houtsnijwerk, eenvoudig in uitvoering maar vol pathos en drama. De Duomo, verbouwd in de 16e eeuw, zou binnenkort weer in zijn oude glorie moeten terugkeren.

    Tegenover de kathedraal staat de achthoekige Doopkapel, vermoedelijk gebouwd op de plek van een heidense tempel. De zwart-wit gestreepte doopkapel valt op door zijn harmonieuze marmeren deuropening en zijn prachtige doopvont, gebeeldhouwd door Sansovino in 1505.
